Culturing LCM (L929 Culture media) for mouse macrophage culture

  • L929 cells produce MCSF and other growth factors. Supernatant from L929 cultures can be used to differentiate macrophages.

Calcein staining of macrophages

  • Labelling cells with calcein AM allows for them to be quantitated using a fluorescent plate reader and is useful for various in vitro assays, including chemotaxis assays

Griess assay for nitrate determination

  • This assay is used for measuring nitric oxide production from macrophages (often used in macrophage polarization studies). Note that this assay must be done in RPMI media, not DMEM.
